Le développement d'une PCR unique amplifiant les espaceurs internes tranSo/its (Internai , Trascribed Spacer : ITS) de l'ADN ribosomal des trypanosomes est une option prometteuse pour trouver un compromis entre un diagnostic sensible et spécifique d'une part et d'autre part un diagnostic accessible pour un coût raisonnable. Notre travail s'intègre dans cette dynamique et a consisté dans un premier temps à l'évaluation des amorces polyspécifiques (Tryp 4R èt Tryp 4S) pour la PCR directe et dans un second temps à l'étude de la PCR-ELISA sur les ITS des trypanosomes. Ainsi, pour la PCR directe, 425 échantillons de terrain dont 311 prélèvements sanguins de bovins (buffy coat) et 114 organes de mouche ont été analysés en PCR monospécifique et en PCR-ITS. Les amorces polyspécifiques ont détecté sur l'ensemble des échantillons 127 signaux positifs soit 29,88 %, par contre les amorces spécifiques. d'espèce ont identifié 168 cas positifs soit 39,53 %. Pour les infections à Trypanosoma vivar, le couple d'amorces Tryp 4R et Tryp 48 totalise 97 cas positifs soit uneprévalence de 22,82 %, tandis que les amorces TVW 1 & 2 ont identifié 51 cas soit 12 %. Avec les amorces TCS 1 & 2,99 cas positifs soit 23,29 % sont rapportés à Trypanosoma congolense type savane, contre seulement 5,41 % représentant 23 échantillons révélés positifs à cette espèce en PCR-ITS. Au regard de ces résultats, la PCR utilisant le couple d'amorces polyspécifiques demeure globalement moins sensible que la PCR monospécifique. Cette faible sensibilité est particulièrement observée dans la détection des trypanosomes du sous genre Nannomonas (T. congolense).
Pour la PCR-ELISA, les sondes oligonucléotidiques spécifiques aux rrs des principales espèces de trypanosomes pathogènes du bétail ont identifié avec une spécificité totale les échantillons d'ADN de référence. Mais, la sensibilité de cette technique sur des échantillons de terrain s'est avérée inférieure à celle observée avec la révélation sur gel d'agarose. Par ailleurs, l'application de la PCR-ELISA dans nos conditions reste limitée par son coût élevé. En effet, le coût de t'analyse d'un échantillon par PCR-ELISA tùt tes ITS a été estimé à 1,8 € (environ 1180 Fcfa) pour 4 espèces de trypanosomes détectées. Ce coût représente plus du double de celui de la PCRITS directe estimé à 0,72 € soit 472,30 Fcfa. Par conséquent, la PCR-ITS direcSte constitue une méthode de choix et à privilégier pour un diagnostic de routine. |
